目的:建立高效液相色谱法测定咪喹莫特乳膏含量的方法。方法:采用TopsilTMC8柱(4.6 mm×200 mm,5μm),检测波长为245 nm,流动相为甲醇-水相(1%三乙胺、0.2%十二烷基硫酸钠溶液,磷酸调节pH值为2.0)(52∶48),柱温为40℃,流速为1.0 mL·min-1。结果:咪喹莫特在20.0~200.0μg·mL-1与峰面积呈良好线性关系,r=0.9999;平均加样回收率为99.98%,RSD为0.99%。结论:该方法测定灵敏、选择性、重现性好,可以用于咪喹莫特乳膏中咪喹莫特的含量测定。
Objective: To establish a method for the determination of imiquimod cream by high performance liquid chromatography. Methods: The detection wavelength was 245 nm and the mobile phase consisted of methanol-water phase (1% triethylamine, 0.2% sodium dodecyl sulfate solution, phosphoric acid, pH = 2.0) (52:48). The column temperature was 40 ℃ and the flow rate was 1.0 mL · min-1. Results: Imiquimod had a good linear relationship with the peak area at 20.0 ~ 200.0μg · mL-1, r = 0.9999. The average recoveries were 99.98% and the RSD was 0.99%. Conclusion: The method is sensitive, selective and reproducible. It can be used for the determination of imiquimod in imiquimod cream.
