目的探讨外周血淋巴细胞泛素mRNA的表达与慢性HBV活动性感染的关系及临床意义。方法96例慢性HBV感染并有炎症活动者,其中慢性乙肝组(40例),活动性肝炎肝硬化组(30例),慢性重型肝炎组(26例),同时设立健康对照组(26例)。采用实时荧光PCR检测外周血淋巴细胞泛素mRNA的表达,以-βactin作为内参。组间差异采用单因素方差分析。结果各组患者外周血淋巴细胞泛素mRNA的表达水平均低于对照组(P=0.00),其中慢性重型肝炎组表达水平最低,活动性肝硬化组次之。结论慢性HBV感染并有炎症活动者均存在外周血淋巴细胞泛素mRNA表达不足,病情越重表达水平越低;外周血淋巴细胞泛素mRNA的低表达可能与疾病的发生发展有关。
Objective To investigate the relationship between peripheral blood lymphocyte ubiquitin mRNA expression and chronic HBV infection and its clinical significance. Methods Ninety-six patients with chronic HBV infection and inflammatory activity were enrolled in this study. Among them, chronic hepatitis B (40 cases), active hepatitis cirrhosis (30 cases), chronic severe hepatitis (26 cases) and healthy control group (26 cases) . Real-time fluorescence PCR was used to detect the expression of ubiquitin mRNA in peripheral blood lymphocytes. Β-actin was used as an internal control. Differences between groups using one-way analysis of variance. Results The expression level of ubiquitin mRNA in peripheral blood lymphocytes in each group was lower than that in the control group (P = 0.00). The expression level of ubiquitin mRNA was the lowest in chronic severe hepatitis group and second in active cirrhosis group. Conclusion The expression of ubiquitin mRNA in peripheral blood lymphocytes in patients with chronic HBV infection and those with inflammatory cytokines is low, and the more severe the disease is, the lower the expression level of ubiquitin mRNA is. The low expression of ubiquitin mRNA in peripheral blood lymphocytes may be related to the occurrence and development of the disease.
