目的 :探讨胃癌组织中c erbB 2和 p5 3基因的表达及其意义。 方法 :应用荧光原位杂交技术对 5 5例胃癌及移行区、正常区组织的常规石蜡包埋标本进行检测。结果 :c erbB 2和p5 3基因表达的阳性率分别为36 36 %和 45 45 % ,移行区为 10 91%和 3 6 4% ,正常粘膜区为 3 6 4%和 0 ,三区间差异均有极显著意义 (P值均 <0 0 1)。在肠型和弥漫型胃癌中 ,c erbB 2阳性率分别为 5 1 6 1%和 16 6 7% ,p5 3阳性率分别为 2 5 81%和70 83% ,其在两型间的差异有显著性意义 (P <0 0 5 )。c erbB 2和 p5 3基因表达与胃癌的组织学分级有关 (P <0 0 5或P <0 0 1) ;c erbB 2基因表达阳性与胃癌的淋巴结内转移有关 (P <0 0 5 )。结论 :c erbB 2基因扩增和p5 3基因突变与胃癌的发生、类型、分化和转移有关 ,其测定可有助于确定肿瘤的生物学行为。
Objective: To investigate the expression and significance of c erbB 2 and p53 gene in gastric cancer. METHODS: Fifty-five specimens of gastric cancer, transitional and normal tissues were examined by fluorescence in situ hybridization. RESULTS: The positive rates of c erbB 2 and p53 gene expression were 364.6% and 445.5%, the transitional regions were 1091% and 36.4%, and the normal mucosal regions were 36.4% and 0. Extremely significant (P values < 0 01). In intestinal and diffuse gastric cancer, the positive rates of c erbB 2 were 516 1% and 166 7%, respectively. The positive rates of p53 were 258 1 and 70 83%, respectively. Significant significance (P < 0 05). The expression of c erbB 2 and p53 was related to the histological grade of gastric cancer (P <0 05 or P <0 01). The positive expression of c erbB 2 was related to the metastasis of lymph nodes in gastric cancer (P <0 05). Conclusion : The amplification of c erbB 2 gene and p53 gene mutation are related to the occurrence, type, differentiation and metastasis of gastric cancer. The determination of c erbB 2 gene can help determine the biological behavior of tumor.
