The authors proposed high pressure liquid chromatography (HPLC) determination of V_A in milk and cheese with an average recovery of 93 ± 6%, repeatability of ± 4% relative standard deviation and a limit of detection of 20 ng of rosin oil (saponified V_A ). The authors compared the results obtained with this method with the conventional colorimetric method, with no significant difference between the two. The colorimetric method can measure up to 6 samples per day and uses a considerable amount of experimental equipment. The HPLC method can measure up to 30 samples per day and the required glassware can be reduced by 1/3. This method is simple, fast, specific, reproducible. Method [Reagents] Petroleum ether (40-60 ° C), pyrogallol, anhydrous sodium sulfate and potassium hydroxide (all Merck analytical grade reagents above), acetonitrile for HPLC (USA).
