目的研究Bcl-2家族的表达同多巴胺诱导的PC12细胞凋亡的相关性,探讨帕金森病(PD)神经元的死亡机制。方法应用免疫组织化学及电镜技术,对Bcl-2家族的表达同多巴胺诱导PC12细胞凋亡的相关性进行分析。结果适当浓度DA可诱导PC12细胞凋亡,凋亡早期伴有Bcl-2及Bax表达量及Bcl-2/Bax比值的增高,后期则出现Bcl-2/Bax比值的下降。结论细胞凋亡参与了PD的发病过程,Bcl-Bax比例的变化同凋亡发生密相关。
Objective To investigate the relationship between the expression of Bcl-2 family and the apoptosis of PC12 cells induced by dopamine and to explore the mechanism of death of Parkinson’s disease (PD) neurons. Methods Immunohistochemistry and electron microscopy were used to analyze the correlation between the expression of Bcl-2 family and the apoptosis of PC12 cells induced by dopamine. Results Apoptosis of PC12 cells was induced by proper concentration of DA. The expression of Bcl-2 and Bax and the ratio of Bcl-2 / Bax increased in the early stage of apoptosis, while the ratio of Bcl-2 / Bax decreased in the later stage. Conclusion Apoptosis is involved in the pathogenesis of PD. The change of Bcl-Bax ratio is closely related to apoptosis.
