目的探讨多囊卵巢综合征(Polycystic ovary syndrome,PCOS)患者取卵日卵泡壁层颗粒细胞中脑源性神经营养因子(Brain-derived neurotrophic factor,BDNF)其高亲和力受体原肌球蛋白受体激酶B(Tyrosine receptor kinase B,Trk B)mRNA在卵泡壁层颗粒细胞的表达,评估BDNF及Trk B与卵母细胞成熟之间的相关性。方法采用实时荧光定量多聚核苷酸链式反应(Quantitative real-time polymerase chain reaction,RT-PCR)方法检测56例PCOS患者和44例排卵正常的不孕患者取卵日卵泡壁层颗粒细胞BDNF及Trk B的表达,初步探讨其与卵母细胞成熟的关系及其临床意义。结果①PCOS组BNDF mRNA表达高于对照组(P<0.05),Trk B mRNA表达低于对照组(P<0.05);②PCOS组获卵数显著高于对照组(P<0.05),而卵母细胞成熟率、受精率和优质胚胎率显著低于对照组(P<0.05);③Trk B与BDNF表达强度呈正相关(r=0.502,P<0.05)。结论①BDNF与Trk B可能共同参与了颗粒细胞的生理功能、卵母细胞的成熟以及早期胚胎的发育;②BDNF-Trk B信号通路障碍可能是促使PCOS患者卵巢内卵泡成熟障碍的主要原因。
Objective To investigate the expression of brain-derived neurotrophic factor (BDNF) in granulosa cells of ovarian follicles in patients with polycystic ovary syndrome (PCOS) and its high affinity receptor tropomyosin receptor The expression of Trk B mRNA in the granulosa cells of the follicular wall was evaluated to evaluate the correlation between BDNF, Trk B and oocyte maturation. Results ① The expression of BNDF mRNA in PCOS group was significantly higher than that in control group (P <0.05), while the expression of Trk B mRNA was lower in control group (P <0.05). ② The number of oocytes in PCOS group was significantly higher than that in control group (P <0.05) The maturation rate, fertilization rate and the rate of high quality embryos were significantly lower than those in the control group (P <0.05). The expression of Trk B and BDNF was positively correlated (r = 0.502, P <0.05). Conclusion ①BDNF and Trk B may participate in the physiological function of granulosa cells, oocyte maturation and early embryo development; ②BDNF-Trk B signaling pathway may be the main reason to promote ovarian follicle maturation in PCOS patients.
