目的 :研究金叶败毒颗粒 (简称中药 )对人巨细胞病毒 (HCMV)感染丝裂素活化蛋白激酶(MAPK) p38信号通路的抑制作用 ,并探讨其治疗HCMV感染的分子机制。 方法 :应用原位杂交和免疫组化方法分别检测中药和更昔洛韦 (GCV)干预HCMV感染的细胞 p38mRNA及 pRb蛋白表达水平 ,观察两种药物对HCMV感染人胚胎成纤维细胞 (HEL)的影响。结果 :两种药物均可抑制HCMV在HEL中的增殖 ,但中药能明显抑制 p38mRNA的水平 ,并使其磷酸化底物 pRb蛋白表达降低 ,而GCV无此作用。 结论 :中药可能通过调节MAPK通路而抑制HCMV基因的表达和复制
Objective: To study the inhibitory effect of Jinyebaidu granules (TCM) on mitogen-activated protein kinase (MAPK) p38 signaling pathway in human cytomegalovirus (HCMV) infection, and to explore its molecular mechanism for the treatment of HCMV infection. METHODS: In situ hybridization and immunohistochemistry were used to detect the expression of p38 mRNA and pRb protein in HCMV-infected cells by Chinese herbs and ganciclovir (GCV) respectively. The effects of two drugs on human embryonic fibroblasts (HEL) infected with HCMV were observed. influences. Results: Both drugs could inhibit the proliferation of HCMV in HEL, but traditional Chinese medicine can significantly inhibit the level of p38 mRNA and decrease the expression of pRb protein, which is a phosphorylated substrate, while GCV has no such effect. Conclusion : Chinese medicine may inhibit the expression and replication of HCMV gene by regulating MAPK pathway
