毛细管电泳-电化学发光法分离检测鲳鱼中的色胺

基于色胺具有增强三联吡啶钌(Ru(bpy)32+)电化学发光信号的特性,本文介绍1种毛细管电泳-电化学发光快速分离测定色胺的新方法。重点考察了检测电位、吡啶钌浓度、检测池中缓冲溶液pH值、进样参数等条件对色胺分离检测的影响。结果表明:在最优条件下检测,电位1.15 V,光电倍增管高压800 V,进样条件10kV×10 s,运行高压16 kV,运行缓冲液为pH 6.0 50 mmol/L磷酸盐缓冲溶液。当二盐酸色胺标准样品的浓度在1~8 mmol/L范围,二盐酸色胺的电化学发光强度与其浓度呈良好的线性关系,其线性方程为I=20.649 c+29.839,相关系数r=0.9996(n=8)。检出限(S/N=3)为0.06 mmol/L。将该方法应用于鲳鱼中色胺的分离检测,结果令人满意。该方法具有分析速度快、分离效果好、灵敏度高、重现性好等特点,适用于色胺的快速检测。 Based on the characteristic of tryptamine to enhance the electrochemiluminescence signal of ruthenium (Ru (bpy) 32+), a new method for the rapid separation of tryptamine by capillary electrophoresis and electrochemiluminescence is described in this paper. The effects of test potential, concentration of pyridine ruthenium, pH value of the buffer solution in the test cell and injection parameters on the detection of tryptamine were mainly studied. The results showed that under the optimal conditions, the potential was 1.15 V, the high voltage of photomultiplier tube was 800 V, the injection condition was 10 kV × 10 s, and the high voltage was operated at 16 kV. The running buffer was pH 6.0 50 mmol / L phosphate buffer solution. When the concentration of tryptamine hydrochloride sample was in the range of 1 ~ 8 mmol / L, the electrochemiluminescence intensity of tryptamine dihydrochloride showed a good linear relationship with its concentration, the linear equation was I = 20.649 c + 29.839, the correlation coefficient r = 0.9996 (n = 8). The detection limit (S / N = 3) was 0.06 mmol / L. The method was applied to the separation of test fish for the detection of serotonin, with satisfactory results. The method has the advantages of fast analysis, good separation effect, high sensitivity and good reproducibility, and is suitable for the rapid detection of tryptamine.
