目的:观察丹参酚酸B盐(SA-B)对单侧输尿管梗阻大鼠肾间质纤维化的影响。方法:将雄性SD大鼠随机分为假手术组(12只)、模型组(12只)和治疗组(12只)。模型组大鼠行单侧输尿管结扎(UUO)建立肾小管间质纤维化模型。治疗组在UUO基础上给予SA-B水溶液(12.5 mg.kg-1)灌胃,各组于造模后第14,21天分2批处死。用苏木精-伊红(HE)染色观察肾组织病理学改变,用免疫组织化学方法观察α平滑肌肌动蛋白(α-SMA)的表达,用逆转录-多聚酶链反应(RT-PCR)方法检测肾组织中角蛋白19 mRNA的表达。结果:SA-B能够显著改善梗阻侧肾脏的病理改变,显著减少肾组织α-SMA的表达,增加角蛋白的表达。结论:SA-B可明显改善UUO所致的大鼠肾间质纤维化,其作用可能是通过阻止小管上皮-成肌纤维细胞转分化和抑制成肌纤维细胞的增生和活化而实现的。
Objective: To observe the effect of salvianolic acid B-salt (SA-B) on renal interstitial fibrosis in rats with unilateral ureteral obstruction. Methods: Male Sprague-Dawley rats were randomly divided into sham-operated group (12), model group (12) and treatment group (12). Rats in the model group were subjected to unilateral ureteral ligation (UUO) to establish a tubular interstitial fibrosis model. The treatment group was given intragastric administration of SA-B aqueous solution (12.5 mg.kg-1) on the basis of UUO, and each group was sacrificed in two batches on the 14th and 21st days after modeling. Hematoxylin-eosin (HE) staining was used to observe the histopathological changes of the kidney. The expression of α-SMA was observed by immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR) was used. The expression of keratin 19 mRNA in renal tissue was detected. Results: SA-B can significantly improve the pathological changes of the obstructed kidney, significantly reduce the expression of α-SMA in the kidney, and increase the expression of keratin. Conclusion: SA-B can significantly improve UUO-induced renal interstitial fibrosis in rats, which may be achieved by preventing the transdifferentiation of tubule epithelium-myoblasts and inhibiting the proliferation and activation of myofibroblasts.
