目前在生殖生物学领域中对早期妊娠子宫内膜蛋白质的变化十分重视,因为这与阐明胚泡着床机理,进而开辟控制生育途径有密切关系。本实验是用不同pH范围(pH3.5~10,5~8和8~9.5)的0.5mm平板聚丙烯酰胺凝胶等电聚焦方法对非孕、妊娠不同天数和妊娠后注射高剂量LHRH—A[(D-丙~6,去甘~(10))LH-RH 乙基胺]兔子宫内膜蛋白质进行分析。结果指出: 1.在pH3.5~10范围内的电泳图谱中,早期妊娠兔子宫内膜存在非孕动物所没有的两种妊娠特有蛋白,它们的pI分别为9.65和7.45。在假孕动物子宫内膜不显示pI为9.65的蛋白带,而pI为7.45的蛋白带只能隐约见到。2.在分辨率更高的pH为8~9.5范围内的电泳图谱中,可见到非孕和假孕子宫内膜所不存在的另一条特异蛋白带,它的pI为9.30。3.用高剂量LHRH-A(0.3mg/头、天)处理妊娠动物,则pI为9.65和7.45的两种妊娠特异蛋白受到明显抑制。特别是注射LHRH-A10天的动物,这两种蛋白带完全消失。根据实验结果,本文将上述妊娠特异蛋白与Schwick于1963年发现的兔子宫液中存在的一种妊娠蛋白即子宫珠蛋白(blastokinin)加以比较和讨论,同时对高剂量LH-RHA的抗生育作用也提出了初步看法。
Currently in the field of reproductive biology on early pregnancy changes in endometrial protein attaches great importance because it clarifies the mechanism of implantation of blastocyst, and then open up ways to control fertility are closely related. This experiment is different pH range (pH3.5 ~ 10,5 ~ 8 and 8 ~ 9.5) 0.5mm plate polyacrylamide gel isoelectric focusing method on non-pregnant, pregnant different days and pregnancy after injection of high-dose LHRH- A [(D-propyl ~ 6, to Gan ~ (10)) LH-RH ethylamine] rabbit endometrial protein analysis. The results showed that: 1. In the electrophoretogram of pH3.5 ~ 10, there were two kinds of gestational unique proteins in non-pregnant animals in early pregnant rabbits with pI of 9.65 and 7.45, respectively. In pseudopregnant animals, the endometrium did not show a protein band with a pI of 9.65, whereas a protein band with a pi of 7.45 could only vaguely see. 2. In the more resolved pH range of 8 to 9.5 in the electrophoretic pattern, you can see the non-pregnant and pseudopregnant endometrium does not exist another specific protein band, its pI was 9.30.3. With high Treatment of pregnant animals with doses of LHRH-A (0.3 mg / head, day) significantly inhibited both pregnancy-specific proteins with pIs of 9.65 and 7.45. In particular, animals injected with LHRH-A for 10 days completely disappeared. According to the experimental results, we compared and discussed the pregnancy-specific protein mentioned above with blastokinin, a kind of pregnancy protein found in Schwann’s rabbit uterine fluid in 1963. Meanwhile, the anti-fertility effect of high-dose LH-RHA Also made a preliminary view.
