目的 应用离体大鼠心脏缺血再灌注模型观察基因重组生长激素( G H)对缺血再灌注心脏泵功能的影响。方法 大鼠随机分为二组,分别每天皮下注射 G H 05 I U/kg( G H 组)或等量生理盐水(对照组),7 d 后进行离体心脏缺血再灌注,分别测定缺血前、后再灌注 10 m in、30 m in 的心脏泵功能指标和冠脉流量的恢复率。结果 G H 组离体心脏缺血前基础水平的左室收缩峰压( L V S P)、收缩期左室压力上升速度峰值(+ dp/dt m ax)、舒张期左室压力下降速度峰值(- dp/dt m ax )和冠脉流量( C F) 与对照组比较呈显著差异( P < 001), 缺血后再灌注10 m in、30 m in 的 L V S P、±dp/dt m ax、 C F 与自身缺血前基础水平比较的恢复百分率明显高于对照组( P <005),而 G H 组心率( H R)的恢复率与对照组比较无显著差异(( P > 005))。结论 提示预先应用 G H 可增强心脏泵功能并改善缺血再灌注心脏泵功能的损伤。
Objective To investigate the effect of recombinant human growth hormone (G H) on cardiac pump function during ischemia-reperfusion in isolated rat heart. Methods The rats were randomly divided into two groups. The rats were injected subcutaneously with G H 05 I U / kg (G H group) or normal saline (control group) daily for 7 days, respectively. Cardiac function and coronary flow recovery were measured at 10 and 30 min after ischemia and reperfusion, respectively. Results The LV systolic pressure (L V S P), the peak systolic left ventricular pressure (+ dp / dt m ax), the peak diastolic left ventricular pressure (- dp / dt m ax) and coronary flow (C F) were significantly different from those in the control group (P <001) The recovery percentage of dp / dt m ax and C F compared with the baseline before ischemia was significantly higher than that of the control group (P <005), while the recovery rate of heart rate (HR) in G H group was not significantly different from that of the control group Differences (P> 005)). Conclusion suggests that pre-application of G H enhances cardiac pump function and improves the impairment of cardiac pump function during ischemia-reperfusion.
