目的 探讨人骨形成蛋白 (rh BMP- 2 )对体内、外人脑胶质瘤 SHG44细胞周期和增殖的影响 .方法 体外培养人脑胶质瘤细胞 ,绘制 rh BMP- 2作用前、后的 SHG44细胞生长曲线 ,MTT法测定 rh BMP- 2对 SHG44细胞的增殖影响 ;以流式细胞仪、电镜和琼脂糖凝胶电泳分别检测其细胞周期、超微结构和 DNA片段改变 ;观察局部注射 rh BMP- 2对裸鼠皮下 SHG44胶质瘤增长的影响 .结果 rh BMP- 2可抑制SHG44细胞生长 ,其 G1期 DNA含量明显增加 ,S期含量减少 (P<0 .0 1) ;流式细胞仪检测有凋亡峰存在 (12 .3% ) ;电镜下见胞核染色质浓缩、边集和核碎裂 ,出现凋亡细胞 ;DNA电泳表现为梯状条带 (DNA ladder) .局部注射 rh BMP- 2后的SHG44胶质瘤生长减慢 ,体积缩小 (P <0 .0 1) .结论 rh BMP- 2可阻止 SHG44细胞从 G1期进入 S期 ,并能在体内、外抑制其增殖 ,该作用可能是通过诱导胶质瘤细胞凋亡实现的
Objective To investigate the effect of rh BMP-2 on the cell cycle and proliferation of human glioma SHG44 cells in vitro and in vivo.Methods Human glioma cells were cultured in vitro and the growth of SHG44 cells was observed before and after rh BMP-2 treatment The cell cycle, ultrastructure and DNA fragments were detected by flow cytometry, electron microscopy and agarose gel electrophoresis. The effects of rh BMP-2 on the proliferation of SHG44 cells were observed by MTT assay. On the growth of SHG44 glioma in nude mice.Results rh BMP-2 could inhibit the growth of SHG44 cells, the content of DNA in G1 phase increased significantly, the content of S phase decreased (P <0.01), the flow cytometry Apoptotic peak (12.3%), apoptotic cells appeared on the chromatin condensation, edge set and nuclear fragmentation under electron microscope, DNA ladder showed DNA ladder.The rh BMP- 2 after SHG44 glioma growth slowed down, the volume was reduced (P <0.01) .Conclusion rh BMP-2 can prevent SHG44 cells from G1 phase S phase, and in vitro and in vivo inhibition of proliferation, the role of Probably by inducing glioma cell apoptosis
