目的 探讨利用改进的反向点杂交方法检测已知细胞因子及肿瘤相关基因在胶质瘤、听神经瘤及脉络丛乳头状瘤中表达及突变的可行性 .方法 分别提取正常脑组织和胶质瘤、听神经瘤、脉络丛乳头状瘤组织中的总 RNA,并反转录为 c DNA,随机引物法标记探针 ,与点于特殊尼龙膜上的已知分子进行反向杂交 .结果 该方法可同时检测多种基因在四种组织中的表达 ,其中 BMP- 6、GH,Sis,m dm 2在四种组织中均有较高水平表达 ,而 BMP- 7,IL- 1,- 11,- 12 ,TGF- α,G- CSF,TPO,Endostatin,nm2 3,raf,p5 3,c- er B2 ,ANG在四种组织中均为低表达 ;其余基因在四种组织中的表达水平有较大的差异 .结论 改进的反向点杂交方法可同时检测多种基因的表达 ,并在不同的组织间进行比较 ,是研究肿瘤基因突变的有效方法
Objective To explore the feasibility of detecting the expression and mutation of known cytokines and tumor related genes in glioma, acoustic neuroma and choroid plexus papilloma by using improved reverse hybridization.Methods The normal brain tissue and glioma , Acoustic neuroma, choroid plexus papilloma tissue total RNA, and reverse transcribed into c DNA, random primers labeled probe, and points in a special nylon membrane known molecules reverse hybridization.Results The method can be The expression of BMP - 6, GH, Sis and mdm 2 in all four tissues was detected at the same time, while BMP - 7, IL - 1, - 11, 12, TGF-α, G-CSF, TPO, Endostatin, nm23, raf, p5 3, c-er B2 and ANG were all low in all four tissues; Large differences.Conclusion The improved reverse hybridization method can detect the expression of multiple genes at the same time and compare them in different tissues and is an effective method to study tumor gene mutation
