目的探讨祛痹方对Ⅱ型胶原诱导的免疫性关节炎(CIA)大鼠的作用及免疫调节机制。方法 SD大鼠72只随机分为空白对照组12只,模型组、雷公藤多苷组、祛痹方高剂量组、祛痹方低剂量组各15只。除空白对照组外其余各组采用尾根部皮下注射Ⅱ型胶原与不完全弗氏佐剂的方法诱导出CIA模型,造模第15天祛痹方高、低剂量组分别给予祛痹方混悬液6.66g/(kg·d)、3.33g/(kg·d),雷公藤多苷组给予雷公藤多苷混悬液9.68mg/(kg·d),空白对照组和模型组大鼠等体积纯净水,各组每天灌胃1次,连续4周。观察各组大鼠关节病理损伤并进行半定量评分,检测血浆中抗Ⅱ型胶原抗体(COL-2)、白细胞介素-1β(IL-1β)、白细胞介素6(IL-6)含量。结果模型组大鼠膝关节病理积分,血浆COL-2、IL-1β、IL-6含量较空白对照组明显升高(P<0.05或P<0.01);与模型组比较,祛痹方高剂量组与雷公藤多苷组大鼠关节病理积分,血浆COL-2、IL-1β、IL-6含量明显降低(P<0.05或P<0.01);祛痹方高剂量组大鼠关节病理积分,血浆COL-2、IL-1β水平亦明显低于雷公藤多苷组和祛痹方低剂量组(P<0.05或P<0.01)。结论高剂量祛痹方显著减轻了CIA大鼠膝关节损伤,其作用机理可能与降低血浆COL-2水平,下调血浆IL-1β、IL-6含量有关。
Objective To investigate the effect of Qubi Recipe on type Ⅱ collagen-induced immune arthritis (CIA) in rats and its immunoregulatory mechanism. Methods Seventy two SD rats were randomly divided into blank control group (n = 12), model group, tripterygium glycosides group, high-dose Qubi Fang group and low-dose Qubi Fang group. Except for the blank control group, CIA model was induced by subcutaneous injection of type Ⅱ collagen and incomplete Freund’s adjuvant in the tail of the rats in the other groups. The CIA model was induced on the 15th day in the model group. Solution, 6.66 g / (kg · d) of liquid and 3.33 g / (kg · d) respectively. The tripterygium glycosides group received 9.68 mg / (kg · d) of tripterygium glycosides suspension, and the blank control group and model group rats Purified water volume, each group gavage 1 times a day for 4 weeks. The pathological changes of articular cartilage of rats in each group were observed and semiquantitatively scored. The levels of COL-2, IL-1β and IL-6 in plasma were detected. Results Compared with the model group, the pathological scores of knee joints, plasma COL-2, IL-1β and IL-6 in model group were significantly higher than those in control group (P <0.05 or P <0.01) (P <0.05 or P <0.01). Compared with the rats in the high dose group of Qu Bi Decoction, the joint pathological scores, the contents of COL-2, IL-1β and IL- Plasma levels of COL-2 and IL-1β were also significantly lower than that of tripterygium glycosides and Qubi Fang (P <0.05 or P <0.01). Conclusion High-dose Qubi Fang significantly reduces the knee joint injury in CIA rats. The mechanism may be related to the decrease of plasma COL-2 level and the decrease of plasma IL-1β and IL-6 levels.
