目的探讨Cyclin D1和hMSH2在结直肠癌组织中的表达及其与结直肠癌临床病理特征之间的关系。方法用免疫组化SP法检测50例结直肠癌组织中Cyclin D1和hMSH2的表达,用Kaplan-Meier法分析Cyclin D1和hMSH2的表达与临床病理特征和预后的相关性。用多因素COX模型分析影响患者预后的相关因素。结果在正常组织、癌旁组织和癌组织中Cyclin D1蛋白表达阳性率分别为0、40%和64%,而hMSH2蛋白表达阳性率分别为10%、52%和86%。在结直肠癌组织中Cyclin D1和hMSH2蛋白表达率明显高于正常组织,差异有统计学意义(P<0.01)。Cyclin D1和hMSH2在结直肠癌中表达与分期和分化程度及2年内复发率有关,差异有统计学意义(P<0.05),而与病理类型无关。多因素分析显示,Cyclin D1和hMSH2(RR值分别为2.405和2.312,P<0.05)的表达是结直肠癌的重要预后因素。结论 Cyclin D1和hMSH2基因突变可能与结直肠癌的发生、发展和预后有一定关系。
Objective To investigate the expression of Cyclin D1 and hMSH2 in colorectal cancer and their relationship with the clinicopathological features of colorectal cancer. Methods The expressions of Cyclin D1 and hMSH2 were detected by immunohistochemical SP method in 50 cases of colorectal cancer. The expressions of Cyclin D1 and hMSH2 were analyzed by Kaplan-Meier method in relation to clinicopathological features and prognosis. Using multivariate COX model to analyze the related factors affecting the prognosis of patients. Results The positive rates of Cyclin D1 protein in normal tissues, adjacent tissues and cancerous tissues were 0, 40% and 64% respectively, while the positive rates of hMSH2 protein were 10%, 52% and 86% respectively. The expressions of Cyclin D1 and hMSH2 in colorectal cancer tissues were significantly higher than those in normal tissues (P <0.01). The expression of Cyclin D1 and hMSH2 in colorectal cancer correlated with the stage and differentiation degree and the recurrence rate in 2 years (P <0.05), but not with pathological types. Multivariate analysis showed that the expressions of Cyclin D1 and hMSH2 (RR of 2.405 and 2.312 respectively, P <0.05) were significant prognostic factors for colorectal cancer. Conclusion The mutations of Cyclin D1 and hMSH2 may be related to the occurrence, development and prognosis of colorectal cancer.
