从喇叭菌子实体中分离纯化得到喇叭菌多糖CCPⅠ,通过高效凝胶渗透色谱法和气相色谱法测定其相对分子质量和单糖组成,分析了它的红外光谱特征和核磁共振图谱。研究表明,喇叭菌多糖CCPⅠ的重均相对分子质量为16270 Da,具有典型的多糖红外吸收,是一种含有α和β型吡喃糖苷键的杂多糖,由岩藻糖、甘露糖、葡萄糖和半乳糖组成,摩尔比为1:2.11:3.13:6.45。糖链中C-6大部分被取代,C-2、C-3或C-4部分被取代。采用体外抗补体活性测定方法和四甲基偶氮唑盐微量酶反应比色法评价了喇叭菌多糖CCPⅠ的抗补体活性和抗肿瘤活性,发现当CCPⅠ浓度为4 mg/mL时,抗补体活性达到70.44%,对人肝癌细胞Hep G2和人体乳腺癌细胞MCF-7的增殖均有明显抑制作用且呈剂量依赖性,当CCPⅠ浓度为1000μg/mL时,对Hep G2和MCF-7抑制率分别达到60.06%和55.80%。
Isolation and purification of the polysaccharide CCP1 from hornbill fruiting body, and its relative molecular mass and monosaccharide composition were determined by high-performance gel permeation chromatography and gas chromatography. Anti-complement activity and anti-tumor activity of polysaccharides CCP1 were evaluated by in vitro anti-complement activity assay and MTT assay. It was found that the anti-complement activity and antitumor activity of CCPⅠ at CCPⅠ concentration of 4 mg / mL, Reached 70.44%. The inhibitory rates of Hep G2 and MCF-7 were significantly inhibited in a dose-dependent manner. When the CCPⅠ concentration was 1000 μg / mL, the inhibitory rates of Hep G2 and MCF-7 were respectively Reaching 60.06% and 55.80%.
