目的:测定无症状的过敏者外周血单个核细胞(PBMC)经特异性过敏原刺激后CD4+IL-10+调节性T细胞数量和IL-10水平,并比较其与过敏性哮喘患者和不过敏健康人的差异。方法:选取无症状的尘螨过敏者24例,急性发作期的尘螨过敏性哮喘患者20例,不过敏的健康人22例,均分离PBMC并CFSE染色,分别在不刺激和1mg/L尘螨抗原(rDer p 1)刺激情况下体外培养7d。流式细胞仪检测各组CD4+IL-10+T细胞/CD4+细胞百分比,分析CD4+IL-10+T细胞分裂指数;ELISA法检测各组细胞培养上清液中IL-10和TGF-β1的水平。结果:①rDer p 1刺激后无症状的尘螨过敏者CD4+IL-10+T细胞/CD4+细胞百分比高于过敏性哮喘患者;无论刺激与否,无症状的尘螨过敏者CD4+IL-10+T细胞/CD4+细胞百分比与不过敏的健康人水平接近。②与不过敏的健康人和过敏性哮喘患者比较,无症状的尘螨过敏者表现为rDer p 1刺激后PBMC高水平的IL-10反应。结论:无症状过敏的发生可能与机体对特异性过敏原刺激产生高水平的IL-10反应有关。
OBJECTIVE: To determine the number of CD4 + IL-10 + regulatory T cells and IL-10 in asymptomatic allergic peripheral blood mononuclear cells (PBMCs) stimulated by specific allergens and to compare them with patients with allergic asthma and with or without Differences in people with allergies.
