目的 研究扇贝 (Placopectamagellanicus)裙边中提取的糖胺聚糖 (SS GAG)是否如肝素一样抑制血管平滑肌细胞 (VSMC)的增殖。方法 采用胎牛血清和碱性成纤维细胞生长因子 (bFGF)所诱导的大鼠胸、腹主动脉VSMC增殖模型 ,通过细胞计数、结晶紫染色及MTT比色法观察SS GAG对VSMC增殖的影响 ,并运用免疫组织化学技术和计算机图像分析系统检测SS GAG对VSMC的增殖细胞核抗原(PCNA)和血小板衍化生长因子 (PDGF)表达的影响。结果 SS GAG 5 0 ,10 0和 2 0 0mg·L- 1对 2 0 %胎牛血清和 5 0 μg·L- 1bFGF所诱导的VSMC增殖均有明显抑制作用 ,并能抑制VSMC增殖时PCNA和PDGF的表达。结论 SS GAG能抑制VSMC的增殖 ,该作用可能是通过抑制PDGF和PCNA的表达而实现的。
Aim To investigate whether glycosaminoglycan (SS GAG) extracted from the skirt of Scallop Plagax alal alanicus inhibits the proliferation of vascular smooth muscle cells (VSMCs) like heparin. Methods The thoracic and abdominal aorta VSMC proliferation induced by bFGF in fetal bovine serum was studied. The effects of SS GAG on the proliferation of VSMC were observed by cell counting, crystal violet staining and MTT colorimetry The effects of SS GAG on the expression of proliferating cell nuclear antigen (PCNA) and platelet-derived growth factor (PDGF) in VSMCs were detected by immunohistochemistry and computerized image analysis system. Results SS GAG 50, 100 and 200 mg · L -1 significantly inhibited the proliferation of VSMCs induced by 20% fetal bovine serum and 50 μg · L-1bFGF, and inhibited the proliferation of VSMCs PDGF expression. Conclusion SS GAG can inhibit the proliferation of VSMC, which may be through inhibiting the expression of PDGF and PCNA.
