目的研究珍珠菜总黄酮苷(ZTF)的抗肿瘤作用及其机制。方法通过体内抑瘤试验,考察了ZTF对小鼠肝癌H22的影响;通过体外抑瘤试验,考察了ZTF对人白血病HL-60、K562细胞增殖的影响。并同时考察了ZTF对H22小鼠实体肿瘤组织PCNA表达的影响。结果ZTF对HL-60及K562细胞有明显的增殖抑制作用,ZTF400mg/kg对肝癌H22实体瘤有明显的抑制作用,抑瘤率在30%以上。ZTF可明显下调小鼠肝癌H22肿瘤组织中PCNA的阳性表达,其抑制强度亦呈剂量依赖性趋势。结论ZTF为珍珠菜的抗肿瘤作用有效部位,在体内外均具有一定的抗肿瘤活性,其机制可能是通过下调肿瘤细胞PCNA的表达,干扰肿瘤细胞DNA的合成。
Objective To study the anti-tumor effect and mechanism of total flavonoid glycosides (ZTF) of Lysimachia chinensis. Methods The effects of ZTF on hepatoma H22 in mice were investigated by antitumor experiments in vivo. The effect of ZTF on the proliferation of human leukemia HL-60 and K562 cells was investigated by in vitro tumor inhibition assay. At the same time, the effects of ZTF on PCNA expression in solid tumor tissues of H22 mice were also investigated. RESULTS: ZTF significantly inhibited the proliferation of HL-60 and K562 cells. ZTF 400 mg/kg had a significant inhibitory effect on H22 solid tumor of hepatocellular carcinoma. The tumor inhibition rate was over 30%. ZTF can significantly down-regulate the positive expression of PCNA in H22 tumor tissue of mice with liver cancer, and its inhibitory intensity also showed a dose-dependent trend. Conclusion ZTF is an effective antitumor component of Lysimachia solani. It has antitumor activity both in vitro and in vivo. The mechanism may be through down-regulating the expression of PCNA in tumor cells and interfering with DNA synthesis of tumor cells.
