目的采用HPLC测定槐角总黄酮水解产物中染料木素的含量。方法色谱柱为Kromasil C18柱(150 mm×4.6 mm,5μm),柱温28℃,流动相为甲醇-水(60∶40),流速1 mL.min-1,检测波长260 nm。结果染料木素0.02~0.20μg与内峰面积的线性关系良好(r=0.9999),平均回收率为100.40%,RSD=0.92%(n=6)。结论所用方法简便、准确、重复性好,可作为控制槐角总黄酮水解产物质量的定量依据。
Objective To determine the content of genistein in the hydrolyzate of total flavonoids of Sophora japonica by HPLC. Methods The column was Kromasil C18 (150 mm × 4.6 mm, 5 μm). The column temperature was 28 ℃. The mobile phase was methanol-water (60:40) with a flow rate of 1 mL · min-1 and the detection wavelength was 260 nm. Results The linear relationship between genistein and the peak area was good (r = 0.9999). The average recovery was 100.40% and the RSD was 0.92% (n = 6). Conclusion The method is simple, accurate and reproducible, and can be used as a quantitative basis for controlling the quality of total flavonoids hydrolyzate of Sophora japonica.
