目的:建立UPLC-MS/MS同时测定苦苣苔科植物中7个苯乙醇苷类成分的方法,分析比较桂林产半蒴苣苔属植物中苯乙醇苷含量差异。方法:采用ACQUITY UPLC BEH C18色谱柱(100 mm×2.1 mm,1.8μm),以0.1%甲酸溶液(A)-0.1%甲酸乙腈溶液(B)为流动相,梯度洗脱,流速为0.6 m L/min;柱温40℃;采用电喷雾离子源,负离子检测方式,得到相应的提取离子流图,以峰面积进行定量。结果:7个苯乙醇苷类成分具有良好的线性关系,r值均大于0.9990,方法精密度、重复性和稳定性的RSD值均小于3.5%,加样回收率在97.1%~103.2%,RSD均小于3.6%。14种半蒴苣苔属植物中7个苯乙醇苷在组成和含量上有一定差异。结论:该方法操作简便、准确、重复性好,对寻找高含量苯乙醇苷类资源植物具有指导意义。
OBJECTIVE: To establish a method for the simultaneous determination of 7 phenylethanoid glycosides in the genus Gesneriaceae by UPLC-MS / MS, and to analyze the differences in the contents of phenylethanoid glycosides among the genus Seneca in Guilin. Methods: The mobile phase was eluted with ACQUITY UPLC BEH C18 column (100 mm × 2.1 mm, 1.8 μm) using 0.1% formic acid solution (A) and 0.1% formic acid in acetonitrile as mobile phase at a flow rate of 0.6 m L / min; column temperature 40 ℃; using electrospray ionization source, negative ion detection mode, get the corresponding extracted ion flow graph, to quantify the peak area. Results: The seven phenylethanoid glycosides showed a good linear relationship with r values greater than 0.9990. The RSDs of precision, repeatability and stability were all less than 3.5% and the recovery rates were between 97.1% and 103.2%. The RSDs Less than 3.6%. There are some differences in the composition and content of 7 phenylethanoid glycosides in 14 species of Semperium. Conclusion: The method is simple, accurate and reproducible. It is instructive to find high content of phenylethanoid glycosides plants.
