目的 体外反搏疗法长期以来被用于治疗缺血性心脑血管疾病 ,迄今体外反搏对动脉粥样硬化斑块本身有无影响仍无直接的证据。本研究采用模拟临床长疗程的体外反搏慢性动物实验方法 ,初步观察长期体外反搏对高脂饲养猪动脉粥样硬化斑块形成的影响。方法 3周龄雄性断乳乳猪 15头 ,随机分组为高脂对照组 6头 ,高脂反搏组 6头 ,普通饲料对照组 3头。高脂饲料组在普通乳猪料中添加胆固醇、猪油、蛋黄粉和食用盐 ,初始含量分别为 2 % ,4 % ,6 %和 1% ,采用分段递增法在 30d后将各成分比例递增至 4 % ,8% ,10 %和 1.5 % ,专用搅拌器混合高脂饲料 ,采用自行设计的猪专用囊套和猪体外反搏专用吊床装置对反搏实验组动物进行体外反搏 1h/d ,5d/周 ,累计反搏 30h ,总实验过程持续近 3个月 ,实验结束后取猪胸腹主动脉纵向剖开并修整固定 ,流水及 70 %乙醇漂洗后经SudanⅢ +Ⅳ染液 37°C振荡染色 ,80 %乙醇脱色分化。内膜面展开固定后经JVCKY F30B 3 CCD彩色图像摄录输入仪采集图像 ,采用德国KONTRONIBAS2 .5全自动图像分析系统 ,进行主动脉内膜面Sudan阳性染色区域的面积百分比分析。统计学处理方法 :直接对Sudan阳性染色百分比进行两组间秩和检验 ;计算Sudan阳性染色百分比的平方根的反正弦值 ,非配对T -检验统计该
