目的探讨细胞周期蛋白Cullin-1(Cul-1)和S期激酶相关蛋白2(Skp2)在鼻咽癌及癌前病变组织中的表达及意义,同时对比它们在鼻咽癌及癌前病变组织中的表达水平,比较二者的分布情况,找出其差异性及相关关系。方法应用免疫组化方法检测Cul-1和Skp2蛋白在20例分化型癌、20例未分化癌和20例鼻咽癌前病变中的表达。结果在20例分化型癌、20例未分化癌和20例鼻咽癌前病变中Cul-1的平均阳性表达率分别为76%、77%、54%;阳性表达值分别为1.63±0.7、1.62±0.68、1.15±0.64,Cul-1在2种鼻咽癌中的表达高于癌前病变,差异有统计学意义(P<0.05),2种鼻咽癌之间无差异;Skp2在20例分化型癌、20例未分化癌和20例鼻咽癌前病变中的平均阳性表达率为32%、41%、33%,平均阳性表达值分别为0.53±0.49、0.82±0.54、0.72±0.43,无统计学差异。对20例分化型鼻咽癌标本Cul-1、Skp2阳性表达值进行相关关系分析,两者相关性不显著。结论 Cul-1参与了鼻咽癌发生、发展,Skp2则可能参与了鼻咽癌的发生过程,它参与的细胞周期调节可能是鼻咽癌发生的早期事件;两种蛋白因子在细胞周期调控中的关系有待进一步验证。
Objective To investigate the expression and significance of Cul-1 and S-kinase-related protein 2 (Skp2) in nasopharyngeal carcinoma (NPC) and precancerous lesions (NPC) In the expression level, compare the distribution of the two to find out the differences and correlations. Methods The expression of Cul-1 and Skp2 protein in 20 cases of differentiated carcinoma, 20 cases of undifferentiated carcinoma and 20 cases of nasopharyngeal precancerous lesions were detected by immunohistochemistry. Results The average positive rates of Cul-1 in 20 cases of differentiated carcinoma, 20 cases of undifferentiated carcinoma and 20 cases of nasopharyngeal precancerous lesions were 76%, 77% and 54%, respectively. The positive expression rates of Cul-1 were 1.63 ± 0.7, 1.62 ± 0.68 and 1.15 ± 0.64, respectively. The expression of Cul-1 in the two nasopharyngeal carcinomas was higher than that in the precancerous lesions (P <0.05), and there was no difference between the two nasopharyngeal carcinomas The average positive expression rates of differentiated carcinoma, 20 undifferentiated carcinoma and 20 nasopharyngeal precancerous lesions were 32%, 41% and 33% respectively. Correlation analysis of the positive expression values of Cul-1 and Skp2 in 20 cases of differentiated nasopharyngeal carcinoma specimens showed no significant correlation. Conclusions Cul-1 is involved in the development and progression of NPC. Skp2 may be involved in the pathogenesis of NPC, and its involvement in cell cycle regulation may be an early event in NPC.
