目的:探讨益气活血中药抑制血管平滑肌细胞(VSM)增殖的分子机制。方法:血管平滑肌细胞被培养在四种培养基中:常规培养基作为对照组,氧化型低密度脂蛋白组,以及在氧化型低密度脂蛋白组基础上的高浓度和低浓度两种兔载药血清培养基组。完成培养及干预后,检测PGC-1、NRF-1(核呼吸因子)和m tTFA(线粒体转录因子)蛋白表达,线粒体功能改变(COX酶活性)及细胞活性。结果:与对照组相比,氧化型低密度脂蛋白组PGC-1、NRF-1、m tTFA蛋白表达、线粒体COX(细胞色素C氧化酶)酶活性及细胞增殖活性均增加(P<0.05)。与氧化型低密度脂蛋白组相比,载药血清组PGC-1、NRF、m tTFA蛋白表达及COX酶活性降低(P<0.05);细胞增殖受到抑制(P<0.05),这种抑制效率呈现药物浓度依赖性。结论:益气活血汤药在人血管平滑肌细胞增生过程中通过改变线粒体功能影响细胞的生长状态。
Objective: To explore the molecular mechanisms of Yiqihuoxue herbs in inhibiting the proliferation of vascular smooth muscle cells (VSM). METHODS: Vascular smooth muscle cells were cultured in four media: conventional media as a control group, oxidized low-density lipoprotein group, and high- and low-concentration rabbits on an oxidized low-density lipoprotein group basis. Drug serum medium group. Upon completion of culture and intervention, PGC-1, NRF-1 (nuclear respiratory factor) and m tTFA (mitochondrial transcription factor) protein expression, mitochondrial function changes (COX enzyme activity) and cell viability were examined. Results: Compared with the control group, the expressions of PGC-1, NRF-1, m tTFA protein, mitochondrial COX (cytochrome C oxidase) activity and cell proliferation activity in oxidized low-density lipoprotein group increased (P<0.05). . Compared with oxidized low-density lipoprotein group, PGC-1, NRF, m tTFA protein expression and COX enzyme activity were decreased in drug-loaded serum group (P<0.05); cell proliferation was inhibited (P<0.05). Drug concentration dependence. Conclusion: Yi Qi Huo Xue Decoction can affect the growth of human vascular smooth muscle cells by altering mitochondrial function.
