目的:对173名东北汉族个体6个miniSTR基因座D10S1248,D14S1434,D22S1045,D4S2364,D2S441andD1S1677的遗传多态性进行分析,获得群体遗传数据。方法:应用荧光标记多重PCR扩增,6个基因座在两个反应体系中进行。310遗传分析仪进行毛细管电泳,分析等位基因片段长度大小,应用数据收集和分析软件收集数据并进而分型。所得数据进行统计学分析。结果:6个miniSTR基因座在东北地区汉族群体中表现出很好的遗传多态性。6个基因座观察到的等位基因大小在67~115bp,观察杂合度在0.728~0.827。6个基因座在中国东北地区汉族群体中的累积个体识别率为0.99999,累计非父排除率为0.99331。结论:6个基因座在东北汉族群体中均具有高度的遗传多态性,可以作为CODIS(联合DNA检索系统)基因座的有效补充,在群体遗传学及器官移植中有一定的应用价值。
OBJECTIVE: To analyze the genetic polymorphisms of six miniSTR loci of D10S1248, D14S1434, D22S1045, D4S2364, D2S441 and D1S1677 in 173 northeast Han Chinese individuals and to obtain population genetic data. Methods: Using fluorescent multiplex PCR amplification, six loci were performed in two reaction systems. 310 Genetic Analyzer Capillary electrophoresis, analysis allele fragment length size, the application of data collection and analysis software to collect data and then type. The data obtained for statistical analysis. Results: Six miniSTR loci showed good genetic polymorphism in Han population of Northeast China. The observed alleles at 6 loci ranged from 67 to 115 bp and the observed heterozygosity ranged from 0.728 to 0.827.6 The cumulative individual recognition rate in Han population of northeast China was 0.99999 and the cumulative non-parent rate was 0.99331 . CONCLUSION: All 6 loci are highly genetic polymorphisms in northeastern Han population, which can be used as an effective supplement to CODIS (Joint DNA Search System) loci. They have a certain value in population genetics and organ transplantation.
