目的研究Sp110基因SNP位点多态性与重庆市汉族人群肺结核发病的关系。方法采用病例-对照研究设计,问卷调查收集肺结核相关危险因素;多种方法检测rs1135791C/T、rs722555A/G、5′-UTRrs11679983A/G、rs28930679C/T、rs9061A/G、rs1063154G/T、rs1047254A/G、rs3948464C/T位点SNP多态性,用SAS软件进行显著性检验,用MDR软件分析与肺结核易患性相关的SNP位点的交互作用。结果 (1)Sp110基因rs1135791CT基因型、rs722555GG基因型、rs11679983GG基因型、rs3948464CC基因型在病例组的频率明显高于对照组;(2)Logistic回归分析rs1135791C/T、rs722555A/G、rs11679983A/G、rs3948464C/T基因型,吸烟史、家庭中有结核病人、人均居住面积及BMI与重庆地区汉族肺结核的易患性有关;(3)Sp110基因rs722555A/G与rs1135791C/T、rs11679983A/G、rs3948464C/T3个位点有交互作用。结论 rs1135791C/T、rs722555A/G、rs11679983A/G、rs3948464C/T可能是重庆市汉族人群肺结核易患的危险因素。
Objective To study the relationship between SNP loci in Sp110 gene and the incidence of pulmonary tuberculosis in Han population in Chongqing. Methods A case-control study was designed and questionnaires were used to collect the risk factors associated with tuberculosis. A variety of methods were used to detect rs1135791C / T, rs722555A / G, 5’-UTRrs11679983A / G, rs28930679C / T, rs9061A / G, rs1063154G / T, rs1047254A / G , Rs3948464C / T SNP polymorphism, using SAS software significant test, using MDR software analysis of tuberculosis susceptibility SNP site interaction. Results (1) The frequencies of rs1135791CT genotype, rs722555GG genotype, rs11679983GG genotype and rs3948464CC genotype of Sp110 gene in case group were significantly higher than those in control group; (2) Logistic regression analysis of rs1135791C / T, rs722555A / G, rs11679983A / (3) Sp110 gene rs722555A / G and rs1135791C / T, rs11679983A / G, rs3948464C / T genotypes, smoking history, tuberculosis patients in the family, living area per capita and BMI were associated with susceptibility to tuberculosis in Han nationality in Chongqing area; T3 sites have interaction. Conclusion rs1135791C / T, rs722555A / G, rs11679983A / G and rs3948464C / T may be susceptible to pulmonary tuberculosis in Han nationality in Chongqing.
