目的研究荔枝果肉乙酸乙酯提取物对巨噬细胞J774分泌促炎递质前列腺素E(2PGE2)和一氧化氮(NO)的影响,初步探讨其诱导炎症性“上火”反应的机制。方法用不同浓度的荔枝乙酸乙酯提取物处理巨噬细胞J774,分别采用紫外分光光度法和Griess试剂法检测PGE2和NO含量;半定量RT-PCR检测环加氧酶(COX-2)和诱导型一氧化氮合酶(iNOS)的表达。结果与空白对照组相比,荔枝乙酸乙酯提取物在100-900μg/ml的浓度范围内可显著提高巨噬细胞J774上清PGE2、NO的含量(P<0.05),900μg/ml提取物能显著提高COX-2和iNOS的mRNA表达水平(P<0.05)。结论荔枝果肉乙酸乙酯提取物能够诱导巨噬细胞J774分泌促炎介质,从而引起炎症性“上火”反应。
Objective To investigate the effect of ethyl acetate extracts from litchi on the secretion of pro-inflammatory mediators prostaglandin E (2PGE2) and nitric oxide (NO) from macrophages J774 and to explore the mechanism of inducing inflammatory “on” reaction . Methods Macrophage J774 cells were treated with different concentrations of ethyl acetate extracts of litchi, PGE2 and NO were detected by UV spectrophotometry and Griess reagent respectively. Cyclooxygenase (COX-2) and induced by semi-quantitative RT-PCR Type nitric oxide synthase (iNOS) expression. Results Compared with the blank control group, ethyl acetate extract of litchi significantly increased the contents of PGE2 and NO (P <0.05) in the macrophage J774 supernatant in the concentration range of 100-900μg / ml, and the extract of 900μg / ml Significantly increased the mRNA expression of COX-2 and iNOS (P <0.05). Conclusion Litchi pulp ethyl acetate extract can induce the secretion of proinflammatory mediators by macrophages J774, resulting in inflammatory “fire” reaction.
