目的:观察醒脑再造胶囊对血管性痴呆大鼠的保护作用。方法:采用永久性结扎双侧颈总动脉结合眼眶放血法制备大鼠VD模型。术后第3天,进行Y迷宫检测,根据结果将手术动物随机分为模型组、甲磺酸双氢麦角碱(5 mg.kg-1.d-1)、醒脑再造胶囊高、中、低(0.4,0.2,0.1 g.kg-1.d-1)剂量组,另有只分离血管不结扎的设为假手术组,每组10只。第4天开始ig给药,5 mL.kg-1,1次/d,给药3周。通过Y迷宫检测其学习记忆能力,HPLC法检测脑组织内的递质含量,激光多普勒血流量仪检测脑血流速度。结果:Y迷宫检测模型组的正确次数显著低于假手术组(P<0.01),阳性药甲磺酸双氢麦角碱组和醒脑再造胶囊高、中剂量组大鼠的正确次数较模型组明显提高(P<0.01或P<0.05);模型组平均脑血流速度223.5mm.s-1,醒脑再造胶囊3个剂量组较模型显著提高;与正常组比较模型组脑组织去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)3种递质含量降低(P<0.01),阳性药组和醒脑再造胶囊3个剂量组均较模型组显著提高(P<0.01或P<0.05)。结论:醒脑再造胶囊能显著改善血管性痴呆大鼠学习记忆能力,提高脑组织内单胺类递质含量和脑血流量。
Objective: To observe the protective effect of Xingnao Capsule on vascular dementia rats. Methods: The rat model of VD was established by permanent ligation of both common carotid arteries and orbital bleed. On the third postoperative day, the Y maze test was performed. According to the results, the animals were randomly divided into model group, dihydroergotamine mesylate (5 mg.kg-1.d-1), Xingnao Capsule high, Low (0.4,0.2,0.1 g.kg-1.d-1) dose group, and another separate blood vessels without ligation set to sham operation group, 10 in each group. Day 4 began ig administration, 5 mL.kg-1, once / d for 3 weeks. The Y maze was used to detect the learning and memory abilities. The content of neurotransmitters in brain tissue was detected by HPLC. The cerebral blood flow velocity was detected by laser Doppler flowmetry. Results: The correct times of Y maze test in model group were significantly lower than those in sham-operation group (P <0.01). The positive numbers of dihydroergotamine mesylate and Xingnao Capsule groups were significantly higher than those in model group (P <0.01 or P <0.05). The mean cerebral blood flow velocity of the model group was 223.5mm.s-1 and the dosage of Xingnao Capsule was significantly higher than that of the model group. Compared with the normal group, the brain tissue noradrenal gland The content of three neurotransmitters (NE, DA and 5-HT) decreased (P <0.01), and the positive drug group and the Xingnao Capsule 3 group were significantly higher than the model group (P <0.01 or P <0.05). Conclusion: Xingnao Capsule can significantly improve the learning and memory abilities of vascular dementia rats and increase the content of monoamine neurotransmitters and cerebral blood flow in the brain.
