[目的]探讨灵芝孢子提取物对Lactacystin诱导PC12细胞损伤的保护作用。[方法]体外培养PC12细胞,建立Lactacystin诱导PC12细胞损伤的模型,观察细胞形态,用CCK-8法检测细胞活力,annexin V-FITC/PI流式细胞法检测细胞调亡。[结果]用不同浓度的灵芝孢子提取物处理PC12细胞时,细胞存活率与对照几乎一致,并未表现出细胞毒性。经20μmol/L的Lactacystin处理24 h后,PC12细胞活力比对照组降低,仅为对照组的63.2%。模型组经不同浓度的灵芝孢子提取物预处理后,细胞活力明显提高,其保护作用随浓度升高而升高。Lactacystin诱导PC12细胞凋亡,处理24 h后细胞凋亡率为48.86%,经灵芝孢子提取物处理后,细胞凋亡率明显降低。[结论Lactacystin能诱PC12细胞凋亡,灵芝孢子提取物能对Lactacystin致PC12细胞损伤有一定的保护作用。
[Objective] To explore the protective effect of Ganoderma lucidum spore extract on Lactacystin-induced PC12 cell injury. [Method] PC12 cells were cultured in vitro, and the model of PC12 cells induced by Lactacystin was established. The morphology of cells was observed. The viability of cells was detected by CCK - 8 assay. The apoptosis of PC12 cells was detected by annexin V-FITC / PI flow cytometry. [Result] When PC12 cells were treated with different concentrations of Ganoderma lucidum spores extract, the cell viability was almost the same as that of the control and did not show cytotoxicity. After treated with 20μmol / L Lactacystin for 24 h, the viability of PC12 cells was lower than that of the control group, only 63.2% of the control group. After being pretreated with different concentrations of Ganoderma lucidum spore extract, the cell viability of the model group was significantly increased, and its protective effect increased with increasing concentration. Lactacystin induced the apoptosis of PC12 cells, and the apoptosis rate was 48.86% after 24 h treatment. The apoptosis rate of PC12 cells treated with Ganoderma lucidum spore extract was significantly decreased. [Conclusion Lactacystin can induce the apoptosis of PC12 cells. Ganoderma lucidum spore extract can protect Lactacystin-induced PC12 cells to a certain extent.
