目的:探讨痛风舒宁片治疗高尿酸血症的药理作用。方法:痛风舒宁片低、中、高剂量(1.1,2.2,4.4 g·kg-1)和别嘌醇片(40 mg·kg-1)ig连续7 d后,建立尿酸致小鼠高尿酸血症模型,观测对血清尿酸的作用;痛风舒宁片低、中、高剂量(0.75,1.50,3.00 g·kg-1)和别嘌醇片(20 mg·kg-1)ig连续7 d,同时建立腺嘌呤+乙胺丁醇致大白鼠高尿酸血症模型,观察对血清尿酸(UA)、尿素氮(BUN)、肌酐(Cre)的作用,测定肾脏指数并进行肾脏病理组织学检查。结果:痛风舒宁片中、高剂量组小鼠血清UA分别为[(124.00±57.33),(56.27±42.24)μmol.L-1],明显低于模型组[(310.50±74.88)μmol.L-1](P<0.05,P<0.01)。痛风舒宁片高剂量组大鼠血清UA为(86.10±32.09)μmol.L-1,明显低于模型组(137.70±43.21)μmol.L-1(P<0.05);痛风舒宁片中、高剂量组Cre水平为[(45.80±8.46),(48.50±6.80)μmol.L-1],明显低于模型组(59.60±13.53)μmol.L-1(P<0.05,P<0.05);痛风舒宁片低、中、高剂量能改善肾实质病变程度(P<0.05,P<0.01,P<0.01)以及降低肾脏指数(P<0.05,P<0.05,P<0.05)。结论:痛风舒宁片有显著的抗高尿酸血症的作用。
Objective: To investigate the pharmacological effects of Gout Recipe in treating hyperuricemia. Methods: Uric acid-induced high uric acid (UA) was induced in mice by low, medium and high dose of Gout (1.1,2.2,4.4 g · kg -1) and allopurine tablets (40 mg · kg -1) The model of hyperlipidemia and hyperuricemia were observed. The effects of low, medium and high doses of gout on Shining tablet (0.75, 1.50, 3.00 g · kg -1) and allopurine tablets (20 mg · kg -1) , And established adenine + ethambutol induced hyperuricemia model in rats. The effects on serum UA, BUN and creatinine were observed. The renal index was measured and renal pathological examination was performed . Results: The serum levels of UA were (124.00 ± 57.33) and (56.27 ± 42.24) μmol.L-1, respectively, significantly lower than those in the model group [(310.50 ± 74.88) μmol·L -1] (P <0.05, P <0.01). The level of serum UA in high-dose gout group (86.10 ± 32.09) μmol.L-1 was significantly lower than that in model group (137.70 ± 43.21) μmol.L-1 (P <0.05) Cre levels in high-dose group were significantly lower than those in model group (59.60 ± 13.53 μmol·L-1, P <0.05, P <0.05) Gout low, medium and high dose of gout can improve renal parenchymal lesions (P <0.05, P <0.01, P <0.01) and reduce the renal index (P <0.05, P <0.05, P <0.05). Conclusion: Gout Shu Ning tablet has a significant anti-hyperuricemia effect.
