研究了植病生防菌成团泛菌(Pantoea agglomerans)的电击转化条件。结果表明受体菌固体培养、生长到对数早期时的细胞转化效果最好;质粒DNA分子量增加 10倍,转化效率降低 100倍;质粒DNA浓度在0.01~1μg/μL范围内其变化与转化频率呈正比,与转化效率成反比;从成团泛菌中提纯的质粒DNA比从大肠杆菌中提纯的相同质粒DNA转化成团泛菌的效率高出30倍5最佳电击条件为场强15kV/cm、电阻200Q、电容25μF,在此条件下受体菌的存活率约为10%。按给出的条件,每微克5kb大小的质粒DNA可以稳定地得到10~5~10~8个转化子。
The electroporation conditions of Pantoea agglomerans were studied. The results showed that the cell transformation of recipient bacteria was best when grown in early logarithmic phase. The molecular weight of plasmid DNA increased 10-fold and the transformation efficiency decreased by 100-fold. The change of plasmid DNA concentration in the range of 0.01-1 μg / μL The conversion frequency is proportional to the conversion efficiency and is inversely proportional to; the plasmid DNA purified from Pantoea agglomerans 30 times higher than that of the same plasmid DNA purified from E. coli 5 the best shock conditions for the field strength 15kV / cm, resistance 200Q, capacitance 25μF, in this condition the survival rate of the recipient bacteria is about 10%. According to the conditions given, 10 ~ 5 ~ 10 ~ 8 transformants can be stably obtained with 5kb plasmid DNA per microgram.
