目的为探讨地塞米松(DXM)对实验性自身免疫性脑脊髓炎(EAE)大鼠临床指标和单核细胞趋化蛋白1(MCP1)mRNA的影响。方法将正常大鼠、完全抗原和百日咳毒素原液免疫后的大鼠免疫后7d,分成:正常大鼠、EAE组和DXM组。DXM组给予DXM腹腔注射,另外两组给予生理盐水注射。取脑和脊髓制成石蜡切片,进行苏木素伊红(HE)染色和MCP1mRNA的原位杂交(ISH),并比较各项临床指标。结果DXM组的发病率、临床评分和MCP1mRNA的阳性细胞数显著性降低(P<0.01)。结论进一步证实DXM的使用可对EAE有治疗作用。
Objective To investigate the effects of dexamethasone (DXM) on the clinical parameters and MCP1 mRNA in experimental autoimmune encephalomyelitis (EAE) rats. Methods The rats immunized with normal rat, complete antigen and pertussis toxin solution were divided into seven groups: normal rats, EAE group and DXM group. DXM group was given DXM intraperitoneal injection, the other two groups were given saline injection. Paraffin sections were made from the brain and spinal cord, and hematoxylin and eosin (HE) staining and in situ hybridization (ISH) of MCP1 mRNA were performed. The clinical parameters were compared. Results The incidence, clinical score and the number of positive cells of MCP1 mRNA in DXM group decreased significantly (P <0.01). Conclusions The use of DXM is further confirmed to have a therapeutic effect on EAE.
