目的研究游离脂肪酸对大鼠肝脏和骨骼肌细胞酪氨酸磷酸酶蛋白量的影响,以探讨游离脂肪酸在肥胖诱发的胰岛素抵抗中的作用机理。方法分离培养Wistar大鼠肝脏和骨骼肌细胞,分别与软脂酸(0.25mmol/L)或油酸(0.125mmol/L)共同孵育6~24小时,应用Western杂交方法检测SHPTP2和PTP1B的蛋白水平。结果肝脏和骨骼肌细胞的PTP1B蛋白量显著增高(P<005),以24小时为著;SHPTP2蛋白量无改变(P>005)。结论游离脂肪酸促进大鼠肝脏和骨骼肌细胞PTP1B表达,提示游离脂肪酸可能通过增高PTP1B蛋白量抑制胰岛素信号传导诱发胰岛素抵抗
Objective To study the effects of free fatty acids on tyrosine phosphatase (TP) protein in rat liver and skeletal muscle cells to explore the mechanism of free fatty acids in obesity-induced insulin resistance. Methods Wistar rat liver and skeletal muscle cells were isolated and cultured, and incubated with palmitic acid (0.25mmol / L) or oleic acid (0.125mmol / L) for 6 ~ 24 hours respectively. Western blotting was used to detect SHPTP2 and PTP1B protein levels. Results The amount of PTP1B protein in liver and skeletal muscle cells was significantly increased (P <005), which was at 24 hours. There was no change in the content of SHPTP2 protein (P> 0.05). Conclusion Free fatty acid (PFA1B) promotes the expression of PTP1B in rat liver and skeletal muscle cells, suggesting that free fatty acids may induce insulin resistance by increasing the amount of PTP1B protein and inhibiting insulin signaling
