目的探讨伤科接骨片促兔下颌骨缺损骨组织修复的可能作用机制。方法健康雄性新西兰兔72只,随机分为正常对照组(24只)、模型对照组(24只)和伤科接骨片组(24只),除正常对照组外其余各组建立下颌骨缺损模型。正常对照组、模型对照组给予正常饲料40g/d,伤科接骨片组给予伤科接骨片饲料40g/d(约含伤科接骨片0.528g)。分别于造模后第7、14、28、56天各处死6只兔,行下颌骨侧位X线片检查后,收集下颌骨缺损处骨组织块,检测环氧合酶-2(COX-2)蛋白、COX-2mRNA表达。结果与正常对照组比较,模型对照组造模后第14、28天兔下颌骨组织COX-2蛋白及COX-2mRNA表达明显升高(P<0.05);与模型对照组比较,伤科接骨片组造模后第14、28、56天兔下颌骨组织COX-2蛋白及COX-2mRNA表达明显升高(P<0.05)。影像学显示,伤科接骨片组较模型对照组兔下颌骨缺损区成骨速度更快,新生骨密度更大。结论伤科接骨片可能通过上调COX-2蛋白及COX-2mRNA表达促进兔下颌骨缺损骨组织的修复。
Objective To investigate the possible mechanism of Shangke jiegu tablet in promoting the repair of mandibular defect in rabbits. Methods 72 healthy male New Zealand rabbits were randomly divided into normal control group (24), model control group (24), and Xueke Jiegu tablet group (24). The mandibular defect models were established except the normal control group . Normal control group, model control group was given normal feed 40g / d, Shangkejiegu tablet group given ShangkexiaoGu tablets feed 40g / d (containing about Discipline 0.528g). Six rabbits were sacrificed on the 7th, 14th, 28th and 56th day respectively after the model establishment. After the lateral mandibular X-ray examination, the bone tissue pieces of the mandible defect were collected and the expression of COX- 2) protein, COX-2 mRNA expression. Results Compared with the normal control group, the expression of COX-2 protein and COX-2 mRNA in the mandibular tissue of the model control group were significantly increased on the 14th and 28th day after model establishment (P <0.05). Compared with the model control group, The expression of COX-2 protein and COX-2 mRNA in rabbit mandibular tissue on the 14th, 28th and 56th day after model establishment were significantly increased (P <0.05). Imaging studies showed that the injury rate of mandibular defects in Shangkejiege tablet group was faster than that of model control group, and the newborn bone density was greater. Conclusion: Shangke Jiege tablet may promote the repair of mandibular defect bone in rabbits by up-regulating the expression of COX-2 protein and COX-2 mRNA.
