用出生4天的SD大鼠睾丸作器官块组织培养,观察不同浓度的咖啡因单独、与腺苷或苯异丙基腺苷协同对睾丸培养块的生殖细胞数量及所有细胞成份DNA合成的影响。结果表明,高浓度的咖啡因(0.6mM)引起培养块的生殖细胞数目及有DNA合成标记的细胞核数减少。高剂量的腺苷(0.3及0.6mM)能抵抗同剂量的咖啡因对生殖细胞数量的影响;而低10倍浓度的腺苷及苯异丙基腺苷则无此作用。但加入以上两种药物都能拮抗高浓度咖啡因所导致睾丸培养块DNA合成的下降。本文讨论了咖啡因及腺苷作用的可能性机制。
The 4-day-old Sprague-Dawley rat testis was used as explants to observe the effects of different concentrations of caffeine alone or in combination with adenosine or phenylisopropyladenosine on the number of germ cells in testis culture and the DNA synthesis of all cell components . The results showed that the high concentration of caffeine (0.6mM) caused the number of germ cells in culture and the number of nuclei labeled with DNA synthesis decreased. High doses of adenosine (0.3 and 0.6 mM) were able to resist the effects of the same dose of caffeine on the number of germ cells; while adenosine and phenylisopropyladenosine at 10 times lower concentrations did not. However, the addition of these two drugs can antagonize the high concentrations of caffeine caused by the testis culture piece of DNA synthesis decreased. This article discusses the possible mechanisms of action of caffeine and adenosine.
