目的 :研究静脉麻醉药硫喷妥钠 (SP)和异丙酚 (Pro)对新生鼠下丘脑视上核 (SO)神经元自发电活动的影响 ,为进一步探讨全身麻醉作用机理打下基础。方法 :应用膜片钳技术 ,观察SP和Pro对SO神经元自发放电活动的变化。结果 :实验发现SP 2 5 μmol/L或Pro 2 0 μmol/L均明显抑制SO神经元的自发放电频率 ,用正常人工脑脊液(ACSF)冲洗 5分钟后其放电频率可恢复至给药前水平。结论 :SP或Pro对下丘脑SO神经元自发电活动均有可逆性的抑制效应 ,提示SO可能是SP或Pro静脉麻醉药在中枢的重要有作用部位
Objective: To study the effects of intravenous anesthetics such as thiopental sodium (SP) and propofol (Proe) on spontaneous activity of hypothalamic supraoptic nucleus (SO) neurons in neonatal rats, and lay a foundation for further exploration of the mechanism of general anesthesia. Methods: Patch clamp technique was used to observe the changes of spontaneous discharge of SO neurons by SP and Pro. Results: SP 2 5 μmol / L or Pro 2 0 μmol / L significantly inhibited the spontaneous firing rate of SO neurons. The frequency of spontaneous discharge of SO neurons after 5 minutes of normal artificial cerebrospinal fluid (CSF) flushing could be restored to pre-administration level. CONCLUSIONS: SP or Pro have reversible inhibitory effects on spontaneous activity of hypothalamic SO neurons, suggesting that SO may be an important active site of SP or Pro intravenous anesthetics in the central nervous system
