通过单因素考察优化了竹叶黄酮的闪式提取工艺,确定最佳工艺条件为:乙醇浓度80%,料液比1:40,组织破碎提取时间40s。粗提物经AB-8大孔吸附树脂纯化后得总黄酮。与乙醇回流法相比,提取时间从3h缩短至40s,总黄酮得率提高了20.2%,其中,异荭草苷得率提高了63.3%。采用DPPH法对两种方法得到的总黄酮进行抗氧化活性比较,闪式提取法所得总黄酮的清除自由基效果优于乙醇回流法所得总黄酮。对其成分研究表明,禾本科植物淡竹(Phyllostachys nigra var.henonis)叶所含的总黄酮主要以异荭草苷、牡荆苷和异牡荆苷为主。闪式提取法是一种简单、高效的提取竹叶黄酮的方法。
The single-factor study optimized the flash extraction process of bamboo leaf flavonoids, and the optimum technological conditions were as follows: ethanol concentration 80%, solid-liquid ratio 1:40 and tissue disruption extraction time 40s. The crude extract was purified by AB-8 macroporous resin to obtain total flavonoids. Compared with the ethanol reflux method, the extraction time was shortened from 3h to 40s, the yield of total flavonoids increased by 20.2%, of which the yield of isoorientin increased by 63.3%. Compared with the antioxidant activity of the total flavonoids obtained by the two methods by the DPPH method, the total flavonoids scavenging free radicals obtained by the flash extraction method are better than the total flavonoids obtained by the ethanol refluxing method. Studies on its components show that the major flavonoids contained in the leaves of Phyllostachys nigra var. Chenonis are mainly isoorientin, vitexin and isovitexin. Flash extraction method is a simple and efficient method of extracting bamboo leaves flavonoids.
