目的:探讨热应激对小鼠Toll样受体4(TLR-4)表达的影响及TLR-4表达与脾脏指数的关系。方法:采用ELISA法检测35℃、42℃热应激小白鼠血液TLR-4蛋白含量,并计算脾脏指数,HE染色、观察其组织病理学变化。结果:在35℃及42℃热应激组均有TLR-4的表达,35℃组2 h达到最高水平,与对照组相比差异显著(P<0.05),然后随应激时间延长而降低,42℃5 h达到最高水平,与对照组差异不显著(P>0.05)。35℃及42℃应激组脾脏指数均较正常组明显降低(P<0.05),组织病理学观察白髓比例减小,脾小体有不同程度的缩小、分布及结构不规则。结论:轻度、短时热应激促进TLR-4的表达;免疫功能的降低与脾脏变化有一定关系。
Objective: To investigate the effect of heat stress on Toll-like receptor 4 (TLR-4) expression in mice and the relationship between TLR-4 expression and spleen index. Methods: The contents of TLR-4 in blood of 35 ℃ and 42 ℃ heat-stressed mice were measured by ELISA. The spleen index was calculated and HE staining was performed to observe the histopathological changes. Results: The expressions of TLR-4 in heat shock group at 35 ℃ and 42 ℃ reached the highest level at 35 ℃ for 2 h, which was significantly different from that in control group (P <0.05), and then decreased with the prolongation of stress time , Reaching the highest level at 42 ℃ for 5 h, but not significantly different from the control group (P> 0.05). The spleen index of the stress group at 35 ℃ and 42 ℃ was significantly lower than that of the normal group (P <0.05). The proportion of the white pulp in the observation of histopathology was decreased and the spleen was reduced, distributed and irregular in structure. Conclusion: Mild and short-term heat stress can promote the expression of TLR-4. The decrease of immune function is related to the change of spleen.
