目的建立同时测定玄麦甘桔颗粒中甘草苷和甘草酸含量的HPLC。方法采用LiChrospher C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-1.2%甲酸溶液为流动相,梯度洗脱,检测波长254 nm。结果甘草酸和甘草苷在0.005~0.5 mg.mL-1内呈良好的线性关系。甘草苷的回归方程为Y=5 735.789 7X-2.702 7,r=0.999 9,平均回收率为99.7%,RSD为1.72%(n=9),甘草酸的回归方程为Y=6 968.182 6X-3.448 4(r=0.999 8),平均回收率为100.8%,RSD为1.31%(n=9)。结论方法操作简便,结果可靠,重复性好,可作为同时测定玄麦甘桔颗粒中甘草苷与甘草酸含量的方法。
Objective To establish a HPLC method for simultaneous determination of glycyrrhizin and glycyrrhizic acid in Xuanmaigan Ganjie Granules. Methods The column was LiChrospher C18 (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-1.2% formic acid as mobile phase with a gradient of 254 nm. Results Glycyrrhizic acid and glycyrrhizin showed a good linear relationship in 0.005-0.5 mg.mL-1. The regression equation of glycyrrhizin was Y = 5 735.789 7X-2.702 7, r = 0.999 9, the average recovery was 99.7% and the RSD was 1.72% (n = 9). The regression equation of glycyrrhizin was Y = 6 968.182 6X-3.448 4 (r = 0.999 8). The average recovery was 100.8% and the RSD was 1.31% (n = 9). Conclusion The method is simple, reliable and reproducible. It can be used as a method for simultaneous determination of glycyrrhizin and glycyrrhizic acid in Xuanmaigan Ganjie granule.
