目的:了解较长期使用雷公藤甲素对正常大鼠血浆促肾上腺皮质激素释放激素(CRH)含量的影响。方法:雄性SD大鼠20只随机分为2组连续灌胃7周,其中空白对照组10只:2%丙二醇2mL·kg-1,日2次;雷公藤甲素组10只:2%丙二醇雷公藤甲素混溶液20μg·kg-1,日2次。用酶联免疫吸附实验检测各组大鼠血浆CRH含量。结果:雷公藤甲素组血浆CRH含量高于空白对照组。结论:雷公藤甲素对下丘脑-垂体-肾上腺轴(HPAA)可能作用机理之一是兴奋下丘脑CRH神经细胞,使下丘脑合成、分泌CRH增加。
Objective: To understand the effect of long-term use of triptolide on plasma corticotropin-releasing hormone (CRH) levels in normal rats. Methods: Twenty male Sprague-Dawley rats were randomly divided into 2 groups. The rats in the blank control group were treated with 2% propylene glycol (2mL · kg-1) twice a day for 2 weeks and 10% triptolide Triptolide mixed solution 20μg · kg-1, 2 times a day. Serum CRH level in each group was detected by enzyme-linked immunosorbent assay. Results: Plasma CRH level in triptolide group was higher than that in blank control group. CONCLUSION: Triptolide is one of the possible mechanisms of action on the hypothalamus-pituitary-adrenal axis (HPAA) by stimulating the hypothalamic CRH neurons and increasing the synthesis and secretion of CRH in the hypothalamus.
