观察辐射对红细胞膜损伤的远后效应。方法应用7.5%聚丙烯酰胺凝胶电泳法、Percol梯度密度离心、NO-2通透速率测定、DIDS通透抑制率测定等方法,分析5例骨髓型急性放射病(ARS)患者受照后第6年红细胞膜蛋白组成与功能。结果辐射对膜蛋白的损伤持续存在,损伤特征为带8蛋白含量减少,带3蛋白阴离子转运功能低下。进一步分析结果显示,ARS年轻红细胞对NO-2通透时间延迟较中、老年细胞变化明显;ARS红细胞对DIDS通透抑制效应下降。结论结果提示,辐射对红细胞膜蛋白的损伤环节在骨髓红细胞合成期;ARS红细胞膜外侧带3结合位点可能发生病变,导致阴离子转运功能下降;同时,ARS红细胞可能存在提前衰老机制
Far-reaching effects of radiation on erythrocyte membrane damage were observed. Methods Five cases of acute myeloid leukemia (ARS) were analyzed with 7.5% polyacrylamide gel electrophoresis, Percol gradient density centrifugation, NO-2 permeation rate and DIDS permeation inhibition rate. Sixth year erythrocyte membrane protein composition and function. Results The damage to membrane protein persisted by radiation, the damage characteristic was that the content of band 8 protein decreased and the band 3 protein anion transport function was low. Further analysis showed that ARS young red blood cells delayed the passage of NO-2 more time than older cells changed significantly; ARS erythrocyte permeability of DIDS decreased. Conclusions The results suggest that the damage of erythrocyte membrane protein by radiation is in the process of erythrocyte synthesis. The 3-binding site on the erythrocyte membrane of ARS may be diseased, leading to the decrease of anion transport function. Meanwhile, ARS may have premature aging mechanism
