目的:探讨喉癌P53基因突变与突变性P53基因蛋白表达之间的相互关系及意义。方法:对40例喉癌新鲜手术标本采用PCR—SSCP检测P53基因突变,免疫组化方法(LSAB)检测突变型P53基因蛋白表达,并比较其相互间的关系。结果:40例喉癌中有26例基因突变阳性(65%),P53基因蛋白表达阳性者23例(57.5%);P53基因突变与其突变型蛋白表达比较,两者皆阳性为45%,两者均阴性为22.5%,其一致率为67.5%(P<0.05)。结论:P53基因突变与其蛋白表达相吻合。两种检测方法在喉癌的分子病理学诊断及其预后方面同样具有重要的价值。
Objective: To investigate the relationship between the mutation of P53 gene and the expression of mutant p53 gene in laryngeal carcinoma. Methods: P53 gene mutation was detected by PCR-SSCP in 40 fresh laryngeal specimens. The expression of mutant P53 protein was detected by immunohistochemistry (LSAB) and the relationship between them was compared. RESULTS: Twenty-six cases of LSCC were positive for mutation (65%) and 23 cases (57.5%) were positive for P53 protein. The positive rate of P53 gene mutation was 45% , Both negative for 22.5%, the consistency rate was 67.5% (P <0.05). Conclusion: P53 gene mutation is consistent with its protein expression. Both detection methods are also of great value in the molecular pathological diagnosis of laryngeal cancer and its prognosis.
