目的对荞麦过敏原的主要致敏蛋白进行研究。方法收集26例荞麦过敏症患者,详细询问临床病史,留取血清建立血清库;荧光免疫分析仪ImmunoCAP系统检测荞麦特异性IgE水平;用免疫印迹法测定每种致敏蛋白组分的灰度值。结果共收集荞麦过敏患者26例(男10例、女16例,平均年龄29岁)。荞麦特异性IgE的中位数水平是8.385 KUA/L。免疫印迹法识别了荞麦过敏原中的15种致敏蛋白:10 kDa(8%)、13kDa(4%)、14 kDa(12%)、16 kDa(4%)、18~19 kDa(100%)、22~24 kDa(46%)、34 kDa(31%)、36 kDa(58%)、38kDa(15%)、40~50 kDa(46%)、54 kDa(23%)、56 kDa(19%)、69~70 kDa(46%)、100~130 kDa(27%)和130~170kDa(19%)。结论荞麦中的18~19、22~24、36、40~50和69~70 kDa蛋白显示高阳性率,可能为荞麦过敏原的主要致敏蛋白。
Objective To study the major allergens of buckwheat allergens. Methods Twenty-six patients with buckwheat hypersensitivity were enrolled. The clinical history was collected and serum was collected to establish a serum pool. Immunohistochemistry was used to detect the specific IgE level of buckwheat. The gray value of each sensitized protein was determined by Western blotting . Results A total of 26 buckwheat allergic patients were collected (10 males and 16 females, mean age 29 years). The median level of buckwheat-specific IgE was 8.385 KUA / L. Immunoblotting identified 15 allergens in buckwheat allergens: 10 kDa (8%), 13 kDa (4%), 14 kDa (12%), 16 kDa (4%), 18-19 kDa (100% ), 22-24 kDa (46%), 34 kDa (31%), 36 kDa (58%), 38 kDa (15%), 40-50 kDa (46%), 54 kDa (23%), 56 kDa 19%), 69-70 kDa (46%), 100-130 kDa (27%) and 130-170 kDa (19%). Conclusion The 18 ~ 19,22 ~ 24,36,40 ~ 50 and 69 ~ 70 kDa proteins in buckwheat show high positive rate, which may be the main allergen of buckwheat allergen.
