目的:研究发酵虫草菌粉对大鼠细胞色素P450酶CYP1A2,CYP3A4,CYP2E1的影响。方法:用Cocktail探针药物法,将W istar大鼠按体重随机分2组,灌胃给予33 g.L-1的虫草菌粉生理盐水混悬液给药组,生理盐水对照组,灌胃体积为8.5 mL.kg-1。诱导10 d后静脉注射给予3种探针药物咖啡因、氨苯砜、氯唑沙宗,股动脉插管取血浆,通过HPLC检测各探针药物的代谢以评价各组CYP1A2,CYP3A4,CYP2E1亚型酶的活性;药动学计算采用DAS 2.0软件完成。结果:给予发酵虫草菌粉组的大鼠,咖啡因、氨苯砜代谢均加快,t1/2分别为(2.90±0.47)h,(4.83±0.60)h;氯唑沙宗t1/2与对照组无显著差异。结论:发酵虫草菌粉对大鼠CYP1A2,CYP3A4有诱导作用,对CYP2E1的作用不明显。
OBJECTIVE: To study the effects of fermented Chinese caterpillar fungus powder on rat cytochrome P450 enzymes CYP1A2, CYP3A4 and CYP2E1. METHODS: The Wistar rats were randomly divided into two groups according to their body weight by Cocktail probe drug treatment. The rats were given intragastric administration of 33 gL-1 Cordyceps militaris suspension to the control group, saline control group, and the volume of gastric perfusion was 8.5 mL.kg-1. After 10 days of induction, three probes, caffeine, dapsone, and chlorzoxazone, were intravenously administered. Plasma from the femoral artery was cannulated. The metabolism of each probe was evaluated by HPLC to evaluate each group of CYP1A2, CYP3A4, and CYP2E1 subtypes. The enzyme activity; pharmacokinetic calculations were performed using DAS 2.0 software. RESULTS: The rats fed with the fermented Chinese caterpillar fungus powder group had faster metabolism of caffeine and dapsone, t1/2 was (2.90±0.47) h, (4.83±0.60) h; chlorzoxazone t 1/2 and control. There were no significant differences in the groups. Conclusion: The fermented Chinese caterpillar fungus powder can induce CYP1A2 and CYP3A4 in rats, but it has no obvious effect on CYP2E1.
