目的探讨五种不同血清量倍比稀释后检测孕妇Ig G型抗A(B)效价积分的差异。方法标记试管1~5排,每排10管,第1排各加生理盐水25μL、第2排各加50μL、第3排各加100μL、第4排各加200μL、第5排各加250μL。在第1~5排第1管分别加入纯化后的孕妇血清25、50、100、200、250μL,进行倍比稀释。在第1排各加与孕妇血清抗体相应的30 m L/L红细胞悬液12.5μL、第2排各加25μL、第3排各加50μL、第4排各加100μL、第5排各加125μL,置37℃孵育30 min;洗涤3次,各管加入多特异性抗人球蛋白试剂1滴,以3400 r/min离心15 s,观察结果。结果五种血清量倍比稀释抗体效价平均积分分别为71、88、91、95、92分;65例孕妇Ig G型抗A(B)效价的异常率分别为30.8%(20/65)、36.9%(24/65)、38.5%(25/65)、40.0%(26/65)、40.0%(26/65)。结论 25μL血清量倍比稀释检测孕妇Ig G型抗A(B)效价积分和异常率明显低于50、100、200、250μL血清量,50μL~250μL血清量之间无显著性差异。
Objective To investigate the differences of Ig G type anti-A (B) titers in pregnant women after being diluted by five different sera. Methods The test tubes were labeled with 1 to 5 rows of 10 tubes in each row. 25μL of saline was added to the first row, 50μL of each of the second row, 100μL of the third row, and 200μL of the fourth row and 250μL of the fifth row. Conclusion The titer and abnormality of Ig G type anti-A (B) in pregnant women were significantly lower than those of 50, 100, 200 and 250 μL serum with 25 μL serum doubling dilution, and there was no significant difference between 50 μL and 250 μL serum.
