目的探讨5一氟脲嘧啶(5-Fu)对靶动脉壁的影响并分析其原因及意义。方法将实验用兔7只中5只行股动脉穿刺给5-Fu注射液,另2只经耳动脉给5-Fu。股动脉给药后7天作X线造影和光镜下病理检查,耳动脉给药后24小时作电镜病理检查。结果5例股动脉造影中4例出现明显狭窄;光镜下表现为较明显内皮细胞脱落、破碎,内弹力膜断裂、不完整或内皮细胞增生明显、呈多层,内弹力纤维聚集,造成管腔狭窄;电镜下见血管内皮细胞膜破裂.胞浆严重空泡变,线粒体结构模糊,内皮下出现以粒细胞为主的白细胞浸润。结论5-Fu注射液对靶动脉壁的刺激和毒性作用导致明显的血管损伤。
Objective To investigate the effect of 5-Fu on the target artery wall and to analyze its cause and significance. Methods Five rabbits in the experiment were implanted with 5-Fu injection in 5 femoral arteries and the other 2 were given 5-Fu via the ear artery. 7 days after administration of femoral artery for X-ray and light microscopy pathology, 24 hours after administration of the ear artery for electron microscopy. Results In 5 cases of femoral artery angiography, 4 cases showed obvious stenosis. Under the light microscope, the endothelial cells were exfoliated, broken, the internal elastic membrane was broken, and the endothelial cell proliferation was incomplete or obvious. The multilayered and elastic fibers gathered, Narrow cavity; under the electron microscope, vascular endothelial cell membrane rupture. Severe cytoplasm vacuolization, fuzzy structure of mitochondria, there are leukocyte infiltration under the endothelium. Conclusion 5-Fu injection on the target artery wall stimulation and toxicity lead to significant vascular damage.
