目的研究甜茶苷对变形链球菌Streptococcus mutans致龋能力的影响。方法将变形链球菌菌液加入甜茶苷后于恒温厌氧环境下培养,测试其pH值变化、黏附率,运用碳氢化合物法测定其表面疏水率。采用Bradford法以Neson-samogyi分别测量总蛋白的量以及还原糖的量,计算葡萄糖基转移酶(glucosyltransferase,GTF)活力,采用Anihrone法测量水不溶性葡聚糖(water-insoluble glucans,WIG)的量。结果加入甜茶苷的菌液pH变化值、黏附率、表面疏水率、GTF活力及WIG的量与对照组之间均有显著性差异(P<0.01)。结论甜茶苷对变形链球菌产酸能力、黏附能力、GTF活力及WIG合成具有显著抑制作用。
Objective To study the effect of resveratrol on cariogenic ability of Streptococcus mutans. Methods Streptococcus mutans were added into the polysaccharides and incubated under constant temperature and anaerobic conditions. The changes of pH value and adhesion rate were measured. The surface hydrophobicity was measured by the hydrocarbon method. Bradford method was used to measure the amount of total protein and the amount of reducing sugar in Neson-samogyi respectively. The activity of glucosyltransferase (GTF) was calculated and the amount of water-insoluble glucans (WIG) was measured by Anihrone method . Results The changes of pH value, adhesion rate, surface hydrophobicity, GTF activity and WIG content of the bacterial cells with addition of resveratrol were significantly different from those of the control group (P <0.01). Conclusion The glycosides have a significant inhibitory effect on the acidogenicity, adhesion, GTF activity and WIG synthesis of Streptococcus mutans.
