目的观察益气逐瘀方参元丹对心肌梗死大鼠心肌损伤标志物及缺血心肌组织miR-24基因表达的影响。方法 50只SD大鼠随机分为假手术组(Sham组)、模型组(Model组)、参元丹高剂量组(H-SYD组)、参元丹中剂量组(MSYD组)、参元丹低剂量组(L-SYD组),每组10只,采用左冠状动脉前降支结扎法复制心肌梗死模型,H-SYD组、M-SYD组、L-SYD组从术后第2天开始参元丹药液灌胃,Sham组和Model组等剂量生理盐水灌胃,共给药2周。治疗结束时,观察各组大鼠血清肌酸激酶(CK)、乳酸脱氢酶(LDH)水平及缺血心肌组织miR-24基因表达情况。结果与Model组比较,H-SYD组、M-SYD组及L-SYD组均能显著降低血清CK、LDH水平(P<0.05,P<0.01),H-SYD组、M-SYD组能够显著升高缺血心肌组织miR-24表达水平(P<0.05,P<0.01)。结论益气逐瘀方参元丹能够减轻心肌梗死大鼠心肌损伤,其作用机制可能与上调缺血心肌组织miR-24基因表达有关。
Objective To observe the effect of Yiqi Zhuyu Fang Shen Dan on myocardial injury markers and the expression of miR-24 in ischemic myocardium in rats with myocardial infarction. Methods Fifty Sprague-Dawley rats were randomly divided into 4 groups: Sham group, Model group, H-SYD group, MSYD group, D-low dose group (L-SYD group), 10 rats in each group. The left anterior descending coronary artery was ligated to establish the model of myocardial infarction. The H-SYD group, M-SYD group and L- At the beginning of the Senate Dan liquid gavage, Sham group and the Model group and other doses of saline gavage for a total of 2 weeks. At the end of treatment, the levels of serum creatine kinase (CK), lactate dehydrogenase (LDH) and the expression of miR-24 in ischemic myocardium were observed. Results Compared with Model group, serum CK and LDH levels were significantly lower in H-SYD group, M-SYD group and L-SYD group (P <0.05, P <0.01), H-SYD group and M-SYD group Increased the expression of miR-24 in ischemic myocardium (P <0.05, P <0.01). Conclusion Yiqi Zhuyu Fang Shen Dan can relieve myocardial injury in rats with myocardial infarction. Its mechanism may be related to the up-regulation of miR-24 gene expression in ischemic myocardium.
