目的 探讨 8 溴 环腺苷酸 (8 Br cAMP)对人膀胱癌BIU 87细胞生长相关基因表达的影响。方法 体外培育的人膀胱癌BIU 87细胞受终浓度为 1× 10 -5mol/L的 8 Br cAMP处理后 ,用RNA斑点印迹技术研究与细胞生长相关的几种基因表达的变化。结果 实验组较对照组c erbB 2基因表达减弱 (478.5 0± 2 2 1.6 4对 14 0 4.0 0± 342 .2 5 ,P <0 .0 0 5 )、H ras基因表达减弱(5 5 9.0 0± 2 6 7.72对 2 36 3 .2 5± 32 4.0 0 ,P <0 .0 1)、突变型 p5 3基因表达减弱 (14 4 4.83± 35 3 .70对36 5 .0± 5 2 .13 ,P <0 .0 2 ) ;而野生型 p5 3基因表达增强 (85 3 .33± 73 .31对 1318.2 5± 35 3 .70 ,P <0 .0 0 1)。结论 8 Br cAMP可通过影响有关信号传递和细胞周期进程基因的表达而抑制细胞生长。
Objective To investigate the effect of 8 Br cAMP on the expression of growth-related genes in human bladder cancer BIU 87 cells. Methods Human bladder cancer BIU-87 cells cultured in vitro were treated with 8 Br cAMP at a final concentration of 1 × 10 -5 mol / L. The changes of several gene expressions related to cell growth were studied by dot blot technique. Results Compared with control group, the expression of c erbB 2 gene in experimental group was weaker (478.5 0 ± 2 2 1.6 4 vs 14 0 4.0 0 ± 342.25, P <0.05), while the expression of H ras gene was weakened (5 5 9.0 0 ± 2 6 7.72 vs 2 36 3 .2 5 ± 32 4.0 0, P <0.01), the expression of mutant p5 3 gene was weakened (14 4 4.83 ± 35 3 .70 vs 36 5 .0 ± 5 2 .13 , P <0. 02), while the expression of wild-type p5 3 gene was enhanced (85.333 ± 73.31 vs. 1318.2 5 ± 35.3 .70, P <0.001). Conclusion 8 Br cAMP inhibits cell growth by affecting the expression of genes involved in signaling and cell cycle progression.
