以筛选减轻抗癌药副作用所致造血功能障碍与免疫功能不全等骨髓功能障碍的植物成分为目的,确立了使用小鼠骨髓细胞的简单的初级筛选方法。运用该方法观察了各种植物提取物的骨髓细胞增殖活性。 将各种植物或生药以水或甲醇提取后,制备提取物溶液。按常规方法从C57BL/6小鼠(雄性,5周龄)的股骨采集骨髓细胞,以α-MEM(含20%FCS、1%BSA、50μM 2-ME)培养液制成浓度为5×10~5个/ml的细胞混悬液,接种在96孔微量培养板或8孔载玻片
A simple primary screening method using mouse bone marrow cells was established for the purpose of screening for plant components that reduce bone marrow dysfunction such as hematopoietic dysfunction and immune dysfunction caused by side effects of anticancer drugs. Using this method, the proliferation activity of bone marrow cells of various plant extracts was observed. After various plants or crude drugs are extracted with water or methanol, an extract solution is prepared. Bone marrow cells were harvested from the femurs of male C57BL/6 mice (male, 5 weeks old) by conventional methods and made into a concentration of 5×10 in α-MEM (containing 20% FCS, 1% BSA, 50 μM 2-ME) medium. ~5 cell suspensions per ml, seeded in 96-well microplates or 8-well slides
